Endonuclease V, also known as deoxyinosine 3′-endonuclease, is a DNA repair enzyme that specifically recognizes deoxyinosine (dI)—the deamination product of deoxyadenosine—in both double-stranded and single-stranded DNA. It cleaves the second phosphodiester bond 3′ to the deoxyinosine, generating a nick with a 3′-hydroxyl and a 5′-phosphate terminus. To a lesser extent, the enzyme also acts on AP sites, urea residues, base-pair mismatches, insertion/deletion mismatches, hairpin structures, unpaired loop regions.
