Phi29 DNA polymerase, derived from Bacillus subtilis phage phi29, possesses both strand displacement activity and strong processive synthesis capability. These properties enable isothermal DNA amplification in vitro without thermal cycling. Phi29 DNA polymerase exhibits 3’→5′ exonuclease activity.
